The production of ambroxide is a classic "nature-to-lab" process, combining the natural origin of plant extraction with the efficiency and consistency of chemical synthesis. The entire process can be divided into two main steps:

Step 1: Extraction and Purification of Sclareol

Clary Sage is commercially grown in multiple regions around the world, such as France, Russia, Bulgaria, and the United States. During the flowering period of plants, their aboveground parts (flowers, stems, leaves) are harvested because the content of sclareol is highest at this time. The raw materials are typically dried after harvesting to lower moisture levels, thereby improving storage stability and preparing them for solvent extraction. After filtering and vacuum distillation to remove the solvent, a semi-solid substance is obtained from the extraction solution. This extract contains sclareol as well as impurities such as wax and pigments. Subsequently, high-purity sclareol crystals were isolated and purified from the extract using purification techniques, including recrystallization.

Step 2: Semi-Synthesis from Sclareol to Ambroxide

This is the core chemical step of converting natural molecules into target spice molecules. The industrial sector has developed multiple mature synthetic routes, among which the most classic and widely used routes include the following key transformations:

  • Oxidative Degradation: High purity Sclareol is subjected to side chain oxidative cleavage using oxidants such as potassium permanganate and chromic acid, resulting in the formation of the key intermediate - Sclareolide. This step is a critical part of the entire synthesis process, significantly influencing both yield and cost.
  • Reduction reaction: The carbonyl group of Sclareolide is reduced by a strong reducing agent (such as lithium aluminum hydride or sodium borohydride) to produce a diol intermediate, Ambrodiol.
  • Dehydration and Cycling: Finally, under the action of an acidic catalyst, the diol intermediate undergoes intramolecular dehydration, forming a five-membered ether ring structure, and ultimately obtaining ambroxide.
  • Final Purification: The crude ambroxide requires further purification through distillation or recrystallization to eliminate by-products and unreacted starting materials, resulting in a commercial-grade product with a purity exceeding 99%.[1]
